The T. reesei cbh2 gene was amplified from first strand cDNA prepared
from T. reesei with primers TRECBH2-L and TRECBH2-R. Sequence analysis
showed that the cloned cbh2 contained three introns. Overlap PCR with
primers CBH2XS-int1, CBH2-int2, CBH2-int3 and TRECBH2-R were used to
remove the native secretion signal and all of the introns.

Khi tách RNA thì có khả năng lẫn DNA, do vậy dùng overlap primer để tránh thu sản phẩm không mong muốn DNA nhiễm tạp này.
