We have a three-year PhD studentship available on the activation of
cryptic
secondary metabolic pathways with the objective to identify and
characterize novel bioactive compounds produced by Streptomyces
ambofaciens.
S. ambofaciens was known to produce two antibiotics: spiramycin, a
macrolide antibacterial agent used in human medicine, and congocidine,
a
pyrrolamide antibiotic. However, the sequencing of the S. ambofaciens
linear chromosome has allowed the identification of 10 putative
secondary
metabolite gene clusters including notably type I and type II PKS, NRPS
and
hybrid PKS-NRPS clusters. We have already shown that one of these
clusters,
the alp cluster, that contains genes encoding a PKS type II highly
homologous to those involved in the biosynthesis of angucycline
antibiotics, is responsible for the synthesis of a novel antibacterial
compound, alpomycin.
The secondary metabolite gene clusters usually includes genes encoding
pathway specific regulation. A part of the project also includes the
understanding of the regulation mechanisms of the secondary metabolites
production. Excellent facilities for this project are available to the
group.
The project is funded by the EC through the 6th framework ActinoGEN
which
involves 15 additional laboratories from across Europe. The successful
candidate will join a group including two postdocs and a technician, as
well as several project students (masters, international exchange,
etc.).
He/she will have the opportunity to participate in different training
events sponsored by ActinoGEN.
Applicants should have good knowledge in molecular biology, microbial
genetics and good communication and organisational skills. The ability
to
design, plan and interpret original experiments is essential.
The deadline for application is the 15th of October. The candidate
should
start no later than January 2007.
