Anh chị giúp em giải thích thuật ngữ "dope oligonucleotide" và inosine. Dope oligonucleotide và inosine này có công dụng gì trong việc tạo site-mutagenesis?
Em cám ơn.

Degenerate primer PCR is in most respects identical to ordinary PCR, but with one major difference. Instead of using specific PCR primers with a given sequence, a mix of PCR primers will be used. Degenerate primer PCR has proven to be a very powerful tool to find "new" genes or gene families. Based on this knowledge, degenerate primer PCR can also be used for mutagenesis. For example, a deoxyinosine triphosphate (dITP) containing primer does not specify the complementary base in the subsequent DNA synthesis. Since all four natural bases may pair with deoxyinosine, there is a 75% probability that any one of the three wrong bases, instead of a correct base, is paired with this ambiguous base. In some cases, trinucleotide phosphoramidite units representing codons for all 20 amino acids can be used to synthesize oligonucleotides. Those random mutagenic oligonucleotides with all possible amino mutations can be obtained quickly
